Abstract:
Hamamelose-diphosphate-U-14C was isolated by ion-exchange chromatography, paper electrophoresis and an enzymatic step from the extract of isolated spinach chloroplasts, which had been allowed to assimilate 14CO2 for 15 min. Since the carbonyl-group of hamamelose-diphosphate was reducible and periodate oxidation yielded 1 mole of formate/mol sugar-diphosphate and no formaldehyde (see table 2), the compound was identified as hamamelose-21, 5-diphosphate. Hamamelose-monophosphate-U-14C was obtained from leaves of Primula clusiana Tausch after photoassimilation of 14CO2 for 15 min. It was concentrated by paper electrophoresis and- chromatography, an enzymatic step, reduction with sodium borohydride and repeated paper chromatography to 70 % purity. Since the other components were known, the elucidation of the structure was possible by the determination of the products of periodate oxidation. Formaldehyde and formate were measured quantitatively whereas glycolic acid was determined qualitatively. The data indicate that hamamelose-monophosphate is a mixture of hamamelose-21-phosphate and -5-phosphate. The occurrence of this mixture is to be expected when free hamamelose arises from hamamelose-21, 5-diphosphate by unspecific dephosphorylation.
